Allah spoke to Moses (peace be upon him) more than once. This included at Mount Tur, as mentioned in Maryam: 52, and at the appointed meeting place, as stated in Surah Al-A'raf: 143. It was reported from Ka'b that Allah spoke to him twice, and from Abu Wail that the speaking occurred many times. Sheikh Ibn Baz mentioned that he had not been informed of a specific number for the times Allah spoke to Moses.
