Does saying "By My Glory" inadvertently, after experiencing a feeling of ecstasy and whisperings, constitute shirk?

What passes through one's mind is not considered shirk (polytheism) nor a sin. As for obsessive thoughts (waswasah), their cure is to turn away from them and pay them no heed. Mixing (of genders) in transportation and other places is a cause for fitna (temptation/strife) and evil. Sitting next to girls is forbidden, and it is obligatory to avoid that. One should stand up if a girl sits next to them, as a precaution against evil and to close the door to fitna.
