The alif after the lam in "Allahu Akbar" is elongated with a natural madd (two vowel counts). It is not permissible to add a waw between the words "Allah" and "Akbar." It is also not permissible to elongate the hamzat al-wasl (connecting hamza) in the word "Allah" because it transforms the statement into a question, and the prayer may be invalidated if the hamza is elongated in the opening takbir.
