The prayer of one who is able to stand behind an Imam who is unable to stand is a matter of scholarly disagreement. The preponderant opinion is that it is not permissible unless he is the Imam of the locality and his illness is one from which recovery is expected. Based on this, the prayer of a wife behind her husband while he is sitting is not valid.
